Description
The National Institutes of Health seeks a contractor to process approximately 600 samples from the Healthy Aging in Neighborhoods of Diversity across the Life Span study. The work includes genomic DNA quantity and quality checks, agarose gel screening, PCR-free library preparation, sequencing on the NovaSeq X-Plus platform at a minimum 30x coverage using 2x150 base-pair runs, and re-sequencing of samples that fail sequencing-based quality control when sample quality and quantity are sufficient. The contractor must perform primary and secondary analysis, including alignment and variant calling with the DRAGEN Germline pipeline, quality-control analyses, joint calling to produce a multi-sample VCF, and confirmation of sample identity, sex, relatedness, contamination, coverage, mapped reads, and other metrics. Deliverables include analytic plates, written pipeline documentation, per-sample CRAMs, gVCFs, VCFs, structural variant and CNV calls, a joint-called multi-sample VCF, a quality-control report and data dictionary, and a sample manifest identifying problems. The contractor must also provide technical consultations and attend Government laboratory staff meetings upon request, and return leftover DNA on dry ice.
