Description
The National Heart, Lung, and Blood Institute seeks non-competitive research support to develop and validate circulating cell-free DNA as a blood-based biomarker of chlorine-induced acute lung injury. The work is intended to establish the mechanistic relationship between chlorine exposure, lung injury, and circulating cell-free DNA and support future diagnostic and triage tools for chemical exposure events. The scope includes use of an established murine chlorine-exposure model; equal numbers of male and female mice; evaluation of acute lung injury at specified post-exposure intervals; bronchoalveolar lavage protein and cell-count analysis; plasma cytokine and chemokine measurements; lung histology and edema assessment; blood-gas testing; epithelial sodium channel and alveolar fluid-clearance studies; hemopexin and heme quantitation; and lung mitochondrial DNA concentration and DNA-lesion analysis.
