The Defense Advanced Research Projects Agency (DARPA), through its Biological Technologies Office (BTO), is actively seeking innovative, post-competitive, and awardable solutions via the ERIS Marketplace. The objective is to develop a new class of rapidly switchable, non-optogenetic protein domains that enable real-time cellular control. This initiative aims to create a robust and versatile molecular tool for massless information transfer across cell membranes, intended to supplement existing synthetic biology platforms. The core problem addressed is the need for precise spatiotemporal control over protein function in advanced synthetic biology systems. While optogenetics is powerful, it has limitations due to potential spectral crosstalk. This effort seeks bio-compatible, modular protein domains, engineered for in vivo expression, that can be regulated by a massless, non-optical external signal (e.g., magnetic fields, focused ultrasound) with sufficient speed and reliability for complex enzymatic machinery. Desired research focuses on achieving activation/deactivation kinetics of 2 seconds, demonstrating at least 300 repetitions in a living cell, and proving the domain's versatility on both split-protein and allosteric configurations. Approaches using magnetogenetics are of high interest, but other innovative modalities meeting performance goals are encouraged. Proposals expanding existing optogenetic, light-based, or chemical systems are explicitly out of scope.
